What exactly does this button do? Does it start hashing all shared files all over again, or does it just refresh its shared files list? I've added/removed from files from the shared folders recently, but they haven't appeared in eMule yet, but I'm sharing so many big files, and it takes so much time and resources to rehash the whole thing, that I'm a little scared to experiment.

That's exactly what you need to do -- press the button ... ...and eMule will update your shared files list -- remove your deleted/moved files and hash newly added files.
